PBS is the default isotonic buffer in cell biology. This is the standard Dulbecco-style formulation at pH 7.4. Quantities below are per litre of 1× PBS.

Method

  1. Dissolve all salts in about 800 mL of deionised water with stirring.
  2. Adjust the pH to 7.4 with HCl or NaOH using a calibrated meter.
  3. Make up to the final volume with deionised water.
  4. Sterilise by autoclaving or filtration; store at room temperature.

Notes & safety

For a 10× PBS stock, multiply each salt by ten and dilute 1:10 before use; note the pH may shift slightly on dilution, so confirm it at 1×.
